HLA-A*32:01 Carriage Is Associated With Lamotrigine-Induced Drug Reaction With Eosinophilia and Systemic Symptoms

This matched case-control genetic association study evaluated HLA risk alleles associated with lamotrigine-induced drug reaction with eosinophilia and systemic symptoms (DRESS) in a US cohort using high-resolution HLA typing and imputation. HLA-A32:01 was strongly associated with increased risk of lamotrigine-induced DRESS, with markedly higher carriage rates in cases versus controls. These findings support the potential utility of pre-prescription HLA-A32:01 screening to reduce the risk of this severe adverse drug reaction.
