In this review, the authors discuss “actinopathies” in terms of the clinical aspects and cellular mechanisms, with a focus on the effect of these mutations on neutrophil function. Several primary immunodeficiencies have been identified in the last 10 years, of which many are caused by mutations in genes related to actin cytoskeleton regulation. These mutations lead to defective function of immune cells, including impairments in motility, signaling, proliferative capacity, and host defense.
